Synchrotron?Based Nano?X?Ray Absorption Near?Edge Structure Revealing Intracellular Heterogeneity of Iron Species in Magnetotactic Bacteria

Magnetotactic bacteria (MTB) sequester iron from the environment to biomineralize magnetite or greigite nanoparticles in magnetosome organelles, though necessity of intracellular storage for formation process is still question. Understanding role would make clear contribution MTB geochemical cycling and its potential importance during biosynthesis application-relevant magnetic nanoparticles. Herein, how scanning X-ray fluorescence microscopy (SXFM) nanoscale absorption near-edge structure (nano-XANES) mapping can spatially chemically identify species reported, creating an opportunity examine biomineralization at single-cell level. Fe K-edge nano-XANES measurements Magnetospirillum gryphiswaldense varied media conditions capacity reveal a significant quantity heterogeneities through distinction between formed magnetosomes material. This component found both early late stages biomineralization. The capabilities providing experimental advantage multidisciplinary field are highlighted.
